雌二醇对豚鼠心室肌细胞L型钙通道的抑制作用
INHIBITORY EFFECTS OF ESTRADIOL ON L-TYPE CALCIUM CURRENT IN GUINEA PIG VENTRICULAR MYOCYTES
【摘要】 目的和方法 :采用全细胞膜片钳技术 ,研究不同浓度 ( 1μmol/L、10 μmol/L、10 0 μmol/L)雌二醇 (estradiol,EST)对豚鼠心室肌细胞L型钙电流 (ICa L)的影响。结果 :①EST明显抑制ICa L,1μmol/L、10 μmol/L、10 0 μmol/LEST分别使峰值ICa L降低 30 .5 % ,5 0 0 % (P <0 .0 1)和 6 8% (P <0 .0 1) ,该作用具有浓度依赖性 ;②EST对ICa L的I V关系曲线未见明显影响 ,其形状和峰值电压保持不变 ;③EST对ICa L的抑制无明显频率依赖关系 ;④EST使通道的激活曲线右移 ,失活曲线左移以及失活恢复时间延长。结论 :EST对豚鼠心室肌细胞L型钙通道具有明显抑制作用
【Abstract】 Aim and Methods: The effect of estradiol (EST) on Ltype calcium current(I Ca.L ) in guinea pig ventricular myocytes was studied by using wholeell patchclamp) technique. Results: EST decreased the I Ca.L significantly in a concentrationdependent manner. At 1 μmol/L, 10 μmol/L and 100 μmol/L of Est the peak current of I Ca.L was depreassed 30.5, 50 and 68 per cent respectively (P< 0.01 ). The IV relationship was not changed and no significant frequencydependent of EST inhibition was observed. EST remarkably shifted activation curve to the right, inactivation curve to the left and prolonged recovery time from inactivation. Conclusion: EST possesses the inhibitory effect on I Ca.L channel in guinea pig ventricular myocytes.
