【摘要】 目的 :检测乳腺癌组织中Fas基因表达并探讨其临床意义。方法 :用流式细胞技术对 36例乳腺癌组织和癌旁乳腺组织中Fas抗原的表达进行检测。同时检测癌组织中细胞凋亡率。结果 :乳腺癌组织 (阳性细胞数 :941± 846 )和癌旁乳腺组织 (阳性细胞数 :5 6 0± 5 5 1)中均有Fas抗原表达 ,其表达水平有统计学差异(P <0 0 5 ) ,细胞凋亡率有显著差异 (P <0 0 5 )。有腋淋巴结转移的乳腺癌Fas抗原表达 (阳性细胞数 :181±5 9)明显低于无腋淋巴结转移者 (阳性细胞数 :132 0± 80 1) ,(P <0 0 5 ) ;乳腺癌组织中雌激素受体的表达与Fas抗原表达及细胞阳性细胞数凋亡率均无关。乳腺癌组织中Fas抗原表达与癌细胞凋亡率有显著相关性 (r =0 6 0 3,P <0 0 1)。结论 :乳腺癌和癌旁乳腺组织均可发生凋亡 ,Fas基因参与乳腺癌的发生和发展。采用特异性改变凋亡阈限的治疗方案 ,可能有利于改变乳腺癌的自然进程

【Abstract】 Purpose:To investigate the apoptosis of breast cancer and its relationship with apoptosis and Fas antigen expression in breast cancer. Methods:Flow immmunocytometry was used to detect the expression of Fas antigen and rate of apoptosis in 36 cases of breast carcinoma tissues and the surrounding non cancerous tissues. Results:Both breast carcinoma tissues (number of positive cells:941±846) and the surrounding noncancerous tissues (number of positive cells:560±551) expressed the Fas antigen,the expression was statistcally significant ( P <0 05), the rate of apoptosis of breast carcinoma (10.1%±7.5%) was significantly higher than that in the surrounding non cancerous tissues (5.6%±5.0%)( P <0 05). Fas antigen expression of breast carcinoma with axillary lymph node involvement (number of positive cells:181±59) was significantly lower than that without axillary lymph node involvement (number of positive cells:1320±801) ( P <0 05). The expression of estrogen receptor in breast carcinoma was not related with the Fas antigen and the rate of apoptosis ( P >0 05). Fas expression in breast carcinoma was associated with the rate of apoptosis (r=0 603, P <0 01). Conclusions:Our study indicates that both in the breast carcinoma tissues and the surrounding noncancerous tissues apoptosis could occur,Fas takes part in the occurence and progression of breast carcinoma. Gene treatment could alter apoptotic abnormality, thus induce apoptosis in cancerous cell expressing Fas antigen.
