苯乙酸对胶质瘤细胞增殖周期的抑制

【摘要】 目的 观察诱导分化剂苯乙酸对胶质瘤细胞C6增殖抑制作用及对细胞周期的影响。方法 细胞计数及 [3H]-TdR掺入法检测苯乙酸对C6细胞的增殖抑制作用。FCM检测苯乙酸对C6细胞周期的影响。结果 C6细胞增殖与苯乙酸剂量及应用时间呈负相关 ,苯乙酸 2 .5,5.0mmol/L组用药第 5d ,对C6细胞的增殖抑制率为 74 .7% ,95.0 % ;第 1d ,3d上述两组用药组G0 /G1期比例分别为 4 5.1% ,56 .2 %和 31.3% ,31.4 %。对照组为 56 .3% ,58.2 %。结论 苯乙酸对C6细胞存在剂量和时间依赖性抑制 ,使细胞周期抑制于G1期。

【Abstract】 Objective To determine the growth-inhibitory activity of phenylacetate (PA) on glioma cell line C6 and to find the influence of the cell cycle by PA. Methods The effect of PA on cell proliferation was determined with [ 3H]-TdR assay, and cell cycle distribution analysis was done with flow cytometry. Results At the dose ranges of 2.5 and 5.0 mmol/L, PA caused a dose-dependent and time-dependent proliferation inhibitory effect. Cells arrested in Phases G0/G1 were observed in PA-treated ( 2.5 , 5.0 mmol/L) cells. Conclusion PA causes a dose-dependent and time-dependent proliferation inhibitory effect, and the main effect of PA is the arrest of glioma cells in Phase G1.
