抗淋球菌单克隆抗体的制备及其生物学特性研究

【摘要】 目的 研制抗淋球菌单克隆抗体 ,为建立新的淋病诊断方法奠定基础。方法 以淋球菌 2 910 6 5株作抗原 ,按常规淋巴细胞杂交瘤技术制备单克隆抗体。并采用ELISA双抗体夹心法研究单抗的特异性和敏感性。结果 建立了 5个持续分泌抗淋球菌单抗的杂交瘤细胞株。它们所分泌的抗体效价可达 1∶8192或更高。 5株单抗均能与淋球菌发生特异性结合 ,但对脑膜炎球菌、葡萄球菌、大肠杆菌等 15种其它抗原不起反应。淋球菌的最小检测量为 1× 10 4 个 /ml。结论 本研究建立的单抗在检测淋球菌时有高度特异性和敏感性。

【Abstract】 Objective To prepare monoclonal antibodies to Neisseria gonorrhoeae for establishing a new diagnostic method of gonorrhoea.Methods Monoclonal antibodies against strain 29106 5 of N.gonorrhoeae were prepared by the lymphocyte hybridoma technipue.The specificity and sensitivity of McAbs were studied using the sandwish method of double antibodies of ELISA.Results five cell lines of hybridoma secreting constantly McAbs to N.gonorrhoeae were obtained.The titer of McAbs reached to 1:8192 or more.All McAbs could react to N.gonorrhoeae,but failed to react to other microbe such as N.Meningitidis、staphylococcus、E.Coliand so on.The minimum dose of N.gonorrhoeae which can be detected by ELISA was 1×10 4cells per ml.Conclusion the sandwich method of ELISA using our McAbs to detect N.gonorrhoeae possessed higher spcificity and sensitivity.
