马立克氏病病毒疫苗CVI988株囊膜糖蛋白gI基因的序列分析
SEQUENCE ANALYSIS OF GLYCOPROTEIN I GENE OF MAREK′S DISEASE VIRUS CVI988
【摘要】 根据马立克氏病病毒 ( MDV)国际标准株 GA株的基因序列设计合成了可扩增 g I基因 ( 1 0 60 bp)的引物 .用 PCR技术 ,以 CVI988基因组为模板 ,扩增得到了预期大小的 PCR产物 .将此 PCR产物和p U C1 8经同样的限制性内切酶 Kpn 和 Bam H 处理后 ,连接、转化、鉴定 ,得到了含有 g I基因的质粒 .将此质粒进行序列分析 ,比较了它与其他强毒和超强毒株的同源性 .用 DNAstar软件对其编码的氨基酸的疏水性和抗原性进行了预测 .
【Abstract】 gI gene coding Glycoprotein was amplified from genomic DNA of MDV CVI988 vaccine strain by PCR. The amplified fragment was cloned into the plasmid vector pUC18 at polylinker via restriction endonuclease KpnⅠ and HindⅢ site. The positive recombinant plasmids with insert were screened by restriction endonuclease analysis and nucleotide sequencing. The obtained gI gene was compared with MDV GA and 648 strains, the results showed that most of the mutations were within the 600 base pairs of N terminal in the open reading frame of gI gene. The hydrophobicity plot and antigenic index of the predicted amino acid were also analysed by softwere DNAstar.
