正常与偏侧帕金森病猴123I-β-CIT SPECT显像研究

【摘要】 目的 评价中枢多巴胺转运蛋白显像剂123I-β-CIT在检测帕金森病中的应用价值。方法 氯胺-T标记法制备123-β-CIT,对正常猴与MPTP诱导的偏侧PD病猴进行SPECT显像研究,并以纹状体/小脑-1作为评价中枢多巴胺转运蛋白丧失程度的指标进行了半定量分析。结果 正常猴全身显像表明,123I-β-CIT脑内摄取约为注射剂量的3%,药物注射后24h SPECT断层显像示正常猴双侧纹状体放射性分布对称,左侧与右侧纹状体/小脑-1值基本一致,偏侧PD病猴右侧(损毁侧)纹状体/小脑值显著低于左侧,提示右侧多巴胺神经终末严重丧失。结论123I-β-CIT是一种良好的显像剂,可用于临床 PD病的早期诊断。

【Abstract】 Purpose To assess the feasibility of 123I -B- CFT imaging in detecting Parkinson’s disease. Methods Normal and MPTP -treated rhesus monkeys underwent spect examination with 123I-B-CIT. The ratio of specific over nondisplaceable binding (striatum/cerebellum- 1) was determined as a measure of the regional density of dopaminergic nerve endings. Results In normal monkey, whole brain activity peaked at 3% injected dose at 1hr after injection. I23I- B- CIT SPECT imaging at 24 hr after injection,normal monkey showing symmetrical tracer distribution in striatiun.the ratios (striatum/cerebellum- 1) had no difference in the left and right,in MPTP- treated monkeys 123I-B- CIT bindings showing a significant decrease in the right in comparison with left. Conclusion The present study demonstrates that 123I - B-GIT could become a useful radio-pharmaceutical for the clinical early dignosis of Parkinson’s disease.
