【摘要】 目的:探讨单纯疱疹病毒胸苷激酶(HXV-TK)基因在人膀胱癌基因治疗中的作用。材料与方法:利用电击基因导入法,将逆转录病毒载体PLNXXTK导入包装细胞PA317,经G418筛选,获得稳定产病毒的PA317/PLNXXTK(PA317/TK)细胞株。采用制备的病毒液转染体外培养的人膀胱癌T24细胞,并联合丙氧鸟苷(GCV)作用,研究其对膀胱癌细胞的生长抑制作用。结果:所获得的PA317/TK细胞能稳定地产生复制缺陷的逆转录病毒载体,滴度为1.65×10~5 cfu/ml。结论:逆转录病毒介导的HXV-TK基因在体外能成功地转染T24细胞,且HXV-TK/GCV系统能明显抑制膀胱癌细胞的生长。这为HXV-TK/GCV在人类膀胱癌治疗方面的进一步体内实验提供了可靠依据。

【Abstract】 : Purpose: In order to investigate the effect about traduction of herpes simplex virous thymidine kinase (HXV-TK) gene to treat human bladder cancer.Method: transduction of human recombinant retrovirus vector-PLNXX-TK, which contained the HXV-TK gene, into the packaging cell PA317 was been done. After selecting by using G418, the HXV-TK gene transduced cells PA317/PLNXXTK was obtained, which could produce replictation-defective retrovirus vector stably. In vitro, the retrovirus mediated HXV-TK gene was transferred into human bladder cancer cells T24 and the growth inhibiting effect by adding different dosage of GCV was observed.Result:The transferred retrovirus packaging cells PA317/PLNXXTK(PA317/TK) were obtained, which could produce replicated defective retrovirus vector and the titre of vector was 1.65×10~5 cfu/ml,and the sensitivity to GCV increased 1000 fold in HXV-TK gene transferred T24 cell compare with the untransfected contrd T24 cells.Conclusion:The retrovirus transferred HXV-TK gene can be transducted into human bladder cancer cell T24. HXV-TK gene transfection with GCV can effectively inhibit growth of human bladder cancer cell T24. The results suggest that therapy of HXV-TK gene will be a potential possibility of treatment of human bladder cancer.
