【摘要】 目的： 用高效液相色谱法测定兔血清中琥乙红霉素的浓度。方法： 采用ＹＷＧ－ Ｃ１８ 色谱分析柱（４－６ ｍ ｍ×１５０ ｍ ｍ ，１０ μｍ）， 岛津ＳＰＤ－ ６ＡＶ 紫外检测器， 甲醇－ ０－０２ ｍｏｌ·Ｌ－ １ － 醋酸铵水溶液（７０∶３０ ， 用氨水调ｐＨ７－３） 为流动相， 红霉素作内标。血液样品在碱性条件下用二氯甲烷提取纯化后进样， 在２１０ ｎｍ 波长处检测。结果： 琥乙红霉素线性范围为０－２５～１２ ｍｇ·Ｌ－ １ （ｒ＝ ０－９９８ ９）， 最低检测量０－０２ μｇ， 最低检出浓度０－０５ ｍｇ·Ｌ－１ ， 平均方法回收率９６－２６％ 。精密度考察琥乙红霉素日内、日间相对标准偏差均小于６％ 。结论： 本方法快速、准确、重复性好。更多还原

【Abstract】 Objective:A HPLC method is described for the analysis of erythromycin ethylsuccinate in rabbit serum.Methods:The YWG-C 18 column(4 6 mm×150 mm,10 μm)was used.The mobile phase consisted of methanol -0 02 mol·L -1 ammonium acetate (70∶30,pH 7 3).Detection was at 210 nm.Results:Erythr omycin was used as achieved by internal standard.The ranges of calibration curves were 0 25～12 mg·L -1 ( r =0 998 9).Detection limits were 0 02 μg and the lowest detectable concentration of roxithromycin was 0 05 mg·L -1 ,while the average recovery of the method was 96 26%.The RSD of intra-day and inter-day assays all less than 6%.Conclusion:The method is quick,precise and good repeat.更多还原
