丙酮沉淀法提取中性β-甘露聚糖酶的条件研究

【摘要】 利用丙酮沉淀提取枯草芽孢杆菌 (Bacillussubtilis)K3- 14发酵产生的中性 β -甘露聚糖酶 ,- 2 0℃时V(发酵液 )∶V(丙酮 )用量为 1∶1.0时 ,提取率为 82 .5 7% ,固体酶制剂酶活力为 19.43万U/g ,1∶2 .0时提取率为 95 .40 % ,所得固体酶净重增加 ,酶活力下降为 8.6 2万U/g ;在 - 2 0℃低温下丙酮对酶的提取影响较小 ,12h内提取率从 97.34 %下降到 87.89% ,下降 9.71% ,而 30℃丙酮对 β -甘露聚糖酶提取影响大 ,从 90 .31%下降到 41.89% ,下降 5 3.6 2 % ,但在 4h以内酶提取率仍能保持在 78.93%以上 ,此工艺在工业上是可行的 .

【Abstract】 Neutral β mannanase produced by Bacillus subtilis K3-14 was extracted by the method of acetone precipitation.The enzyme extraction rate was 82.57% when the fermented both and acetone (-20℃) at the ratio of 1∶1.0,and the enzyme activity was 19.43×10 4?U/g.When the ratio of fermented both and acetone was 1∶2.0,the enzyme extraction rate was 95.40%,so the amount of dry enzyme increased,but the enzyme activity was dropped to 8.62×10 4?U/g,Under the lower temperature (-20℃),there was a little influence of acetone on the enzyme extraction,the extraction rate just dropped from 97.34% to 87.89% within 12?h (dropped 9.71%).However,acetone has a great influence on the enzyme extraction rate at the temperature of 30℃,in which the enzyme extraction rate dropped from 90.31% to 41.89%(dropped 53.62%),but the enzyme extraction rate could still be maintain above 78.93% within 4h at above condition,so we recommend this extraction technique a practicable method in the industrial production.
