【摘要】 从甲型肝炎病毒 (HAV) 2个不同株 (H2 株和L8株 )感染的KMB17细胞中提取总RNA ,经RT -PCR特异性扩增出此 2毒株病毒蛋白酶 2A基因 ,将 2个 2A基因克隆到pGEM -T载体上 ,经DNA序列测定 ,得到2个 2A基因的核苷酸序列 ,序列比较发现它们间及与HAV野毒株 (HM175)相对应序列有突变存在 ,H2 株与HM175相比有一个点突变 ,同源性为 99.72 % ;与HM175比较 ,H2 株和L8株第 3197位核苷酸均由A突变为G ,相应的氨基酸由丝氨酸 (Ser)变为天冬氨酸 (Asp) ;L8株与HM175株相比有 3个点突变 ,同源性为 99.37% ,相应氨基酸均有变化 ;L8株与H2 株相比同源性为 99.6 5 % ,2A区域的变异可能与病毒的生长特性相关 .更多还原

【Abstract】 The genes encoding viral protein enzyme 2A of two hepatitis A virus (HAV) strains (H 2 and L 8) were cloned with RT PCR with specific primers for 2A.The genes were sequenced after inserted into the vector pGEM-T.Sequence comparisons between the two strains and wild strain HM 175 showed that although there were mutations in 2A genes,similarity rates were high,99.37% between L 8 and HM 175 strain,99.72% between H 2 and HM 175 strain,99.65% between L 8 and H 2 strain.The effect of mutations in 2A on replication of HAV strains (H 2 and L 8) was also discussed.更多还原
