5-氟尿嘧啶抑制兔晶状体上皮细胞增殖及眼内毒性的研究

【摘要】 目的 研究 5 氟尿嘧啶 ( 5 Fu)对兔晶状体上皮细胞 (LECs)增殖的影响及眼内毒性。方法 传代兔LECs添加不同质量浓度的 5 Fu ,作用一定时间后 ,MTT法测定OD值。选取对LECs增殖有明显抑制效果的质量浓度 ,进行兔眼前房注射 ,4天后进行角膜内皮分析和眼内炎性细胞分析。结果 体外实验质量浓度为 0 2 5 μg/ml有显著的抑制效果(P <0 0 5 ) ;前房注射的 5 Fu达 5mg/ml时 ,表现出角膜内皮形态改变及前节炎性反应 ,随质量浓度升高 ,程度加重。结论  5 Fu对兔LECs增殖的体外抑制呈时间依赖性和质量浓度依赖性 ,前房注射 5 Fu产生的眼毒性与其质量浓度正相关。

【Abstract】 ObjectiveTo study the anti proliferation effects of 5 fluorouracil(5 Fu) on cultured rabbit lens epithelial cells (LECs) and its intraocular toxicity.MethodsRabbits’ LECs(3×10 4/ml) of 2~5 passages were put into 96 well culture plates.0 125 250?μg/ml of 5 Fu was added.The culture plates were divided into three groups according to exposure time:A (70?min),B(1?d),C(continuous exposure until fussion of cells appeared in the control group).After different exposure period,MTT assay was used to test the OD values.Three effective concentrations of 5 Fu in vitro were selected to be injected into anterior chambers of white rabbits.Pathological slides of the eye balls were made 4?d later.Analysis of corneal endothelial cells and inflammatory cells counting at the corneal margin were performed.ResultsUnder suitable situation,0 25?μg/ml 5 Fu significantly inhibited the proliferation of rabbit LECs in vitro.When injected into the anterior chamber,5?mg/ml 5 Fu induced the morphololgical changes of corneal stroma,iris and ciliary body.ConclusionsThe inhibition on LECs proliferation of rabbits by 5 Fu is time and dose dependent.Under suitable situation,a concentration of 0 125?μg/ml can cause obvious inhibitory effects.The intraocular toxicity of 5 Fu through anterior chamber injection showed positive relation to the concentration.
