碘对甲状腺冷热结节细胞生长的调节特点
The inhibitory effect of iodide on growth of cultured thyrocytes from thyroid adenomas
【摘要】 目的 研究碘对以冷或热结节为表现的甲状腺腺瘤细胞生长的调节特征 ,探讨碘在甲状腺结节发病机理中的作用。方法 手术切取甲状腺腺瘤及腺瘤旁正常组织 ,采用单层细胞培养技术和非同位素标记的细胞增殖分析法 ,检测不同浓度碘化钠 (NaI)经过不同作用时间对甲状腺细胞生长的影响。结果 NaI剂量与时间依赖性地抑制正常与腺瘤甲状腺细胞的增殖 ,其抑制率表现为热结节腺瘤 >正常组织 >冷结节腺瘤 ,且同一类型的甲状腺细胞对NaI的反应性亦存在较大差异。结论 碘对甲状腺生长的调节依赖于细胞的摄碘能力。碘转运体含量的变化在甲状腺结节的发生发展中发挥重要作用。
【Abstract】 Objective To investigate the different response of functioning(hot nodules)and non functioning(cold nodules) thyroid adenomas to iodide.Methods Cell titer 96 Non radioactive Cell Proliferation Assay was used to study the growth of cultured thyrocytes from thyroid adenomas and paraadenomatous tissues in response to various concentration of sodium iodide(NaI).Results NaI inhibited the growth of thyroid cells from both normal and adenomatous tissues in a dose and time dependent manner.The inhibitory effect was greatest in functioning adenomas.Non functioning adenomatous thyroid cells had a lower response than paraadenomatous normal thyrocytes.Moreover,the responsiveness of thyroid cells to iodide on the same type of thyroid was quite different.Conclusion The inhibitory effect of iodide on thyroid growth depends on the iodide uptake by thyrocytes.The changes of expression of sodium/iodide symporter might play an important role in the pathogenesis of thyroid nodules.
