表达β-1,3-葡聚糖酶及几丁质酶基因的转基因烟草及其抗真菌病的研究

【摘要】 利用烟草碱性β-1,3-葡聚糖酶及菜豆碱性几丁质酶基因构建了组成型表达的双价植物 表达载体pBLGC,利用农杆菌介导法转化了烟草,并得到了转基因植株。对其进行分子生物 学分析的结果表明,部分转基因植株在所有检测中都显示较强的阳性反应,这说明外源基因 已整合到烟草基因组中并得到正确表达。活体接菌实验初步表明,转基因植株与对照相比,对 赤星病的侵染具有较强的抵抗能力。

【Abstract】 Plant bivalent expression vector PBLGC for basic tobacco β-1, 3-glucanase and beau chitinase genes were constructed, and the tobacco tissue was transformed. Transgenic tobacco plants were camed out for analysis by PCR, PCR-Southern blot Dot blob Western blot, etc., and the results showed that some transgenic plants gave strong positive signal, it suggested that foreign genes have been integreted into tobacco genome, and expressed correctly. Fungal challenge with Alternaria alternata showed that transgenic plants were much more resistant to this pathogenic fungus.
