淫羊藿苷及济南假单胞菌制剂联合应用体外抗转移的研究

【摘要】 目的 将淫羊藿苷 (icariin ,ICA )和济南假单胞菌制剂 (pseudomonasjinanensisagent ,PJA)作用于人高转移肺癌细胞PG ,从肿瘤转移抑制的多个方面和环节探讨两药抗转移作用的机制 ,并观察其联合用药的作用性质。方法 采用粘附实验、运动侵袭实验、流式细胞术、细胞免疫组化等多种方法进行了检测 ,并按金正均的Q值法计算联合用药效果。结果 ICA、PJA可明显降低PG细胞对层粘连蛋白基质的粘附率 (P <0 0 5 ,P <0 0 5 ) ,并且这种粘附抑制作用有时间依赖性 ;经ICA 2 0 0mg·L-1、PJA 0 35mg·L-1及ICA 2 0 0mg·L-1+PJA 0 35mg·L-1处理 48h后 ,侵袭细胞数由对照组的 470± 5 8分别降为 2 37± 31(P <0 0 1)、316± 34(P <0 0 5 )、199± 2 3(P <0 0 1) ;运动细胞数由对照组的 5 2 0±5 6 ,分别降为 35 4± 5 5 (P <0 0 5 )、375± 19(P <0 0 5 )、2 5 6± 2 2 (P <0 0 1) ,并同时伴有细胞表面粘附分子CD4 4V6、LN R及胞浆内CK18表达的减少。其中两药联合处理对粘附、运动、侵袭能力的抑制作用均为单纯相加效果。药物处理 96h后 ,细胞表面HLA ABC抗原表达率由对照组的10 78%分别升高至 37 5 4%、38 88%、5 0 75 % (P <0 0 5 )。结论 ICA、PJA通过对肿瘤转移多个步骤的抑制而发挥抗转移作用?

【Abstract】 AIM Mechanism of icariin (ICA) and pseudomonas jinanensis agent (PJA) on anti-metasta-sis of a highly metastatic human lung tumor cell line (PG) and the effect type of the combination of ICA,PJA were studied. METHODS PG cells were treated with ICA、PJA, or the combination of both in vitro. Adhesion assay, invasion assay, migration assay, flow cytometry (FCM) and immunohistochemical straining were used. The effects of ICA and PJA combination were evaluated with Q method of Jin Zheng-Jun. RESULTS ICA、PJA could decrease PG cells adhesive ratio to laminin substrate in a time-dependent manner ( P <0 05, P <0 05). The treatment of PG cells with ICA 200 mg·L -1 、PJA 0 35 mg·L -1 、ICA 200 mg·L -1 +PJA 0 35 mg·L -1 for 48 h could decrease the invasive cell numbers from 470±58 to 237±31( P <0 01)、316±34( P <0 05)、199±23( P <0 01). Meanwhile migrative cell numbers decreased from 520±56 to 354±55( P <0 05)、375±19( P <0 05)、256±22( P <0 01) and the expression of CD 44 V 6、LN-R、CK18 of PG cells also reduced. There were enhanced effects on the treatment of ICA and PJA combination. On the other hand, HLA-ABC antigen expression ratio of PG cells increased from 10 78% to 37 54%、 38 88%、 50 75% ( P <0 05) after treatment with ICA 、PJA 、ICA+PJA for 96 h. CONCLUSION ICA、 PJA may play anti-metastatic roles in many aspects and the type of the combination of ICA and PJA is single enhanced effect.
