糖皮质激素对人视交叉上核加压素mRNA表达的影响

【摘要】 目的 观察长期接受糖皮质激素治疗 ,对于人视交叉上核 (SCN)加压素mRNA表达的影响。方法 采用的脑组织为福尔马林固定 ,石蜡包埋标本。切片厚度 :6 μm。SCN连续切片每隔 30 0 μm取片一张 ,用原位杂交的方法 ,显示该切片上加压素mRNA的表达。采用IBAS KAT图像分析系统定量分析SCN加压素mRNA表达信号。结果 实验组SCN加压素mRNA的表达量与对照组相比明显降低 (分别为 44 33± 16 2 8,990 9± 70 4,P =0 0 2 )。实验组SCN表达加压素mRNA的细胞总数与对照组相比亦降低 47% (分别为 1330 6± 36 5 9、2 4939± 1393,P =0 0 3)。在对照组或实验组 ,加压素mRNA总量与脑重量和死后延搁时间均无显著性相关。结论 服用糖皮质激素抑制SCN加压素mRNA的表达 ,可能是患者在接受激素治疗过程中出现睡眠紊乱的生物学基础

【Abstract】 AIM The expression of vasopressin(AVP) mRNA in the suprachiasmatic nucleus (SCN) was observed in patients with and without glucocorticoid treatment. METHODS Formalin fixed and paraffin embeded brain tissue were studied from the patients who had been exposed to glucocorticoids during the last premortem periods. In situ hybridization was performed on every 50th section of the SCN. For quantitative analysis of the in situ signal of the AVP mRNA in the SCN, an IBAS KAT image analysis system was connected to a SONY XL 77CE video camera mounted on a zeiss microscope. Total mask of the silver grains in the profiles was calculated as an estimate of total amount of AVP mRNA. RESULTS The total mask of silver grains in cells was reduced 56% in glucocorticoid exposed patients ( 4 433 ± 1 628 ) as compared with controls( 9 909 ±704) ( P =0 02). There was also a 47% decrease in the total number of profiles in the SCN that expressed AVP mRNA in glucocorticoid exposed patients compared with those in controls( 13 306 ± 3 659 and 24 939 ± 1 393 respectively, P =0 03). In controls, no significant difference was found with aging. No significant correlation was found between amount of AVP mRNA in controls or glucocorticoid exposed group on one hand and brain weight ( r=0 02, P=0 96; r=0 69, P=0 06 , respectively), postmoterdem delay ( r=0 04, P=0 914; r=-0 36, P=0 33 ; respectively) on the other hand. CONCLUSION The mechanism responsible for the sleep disturbances in glucocorticoid exposed patients may be that AVP mRNA expression in SCN is suppressed by glucocorticoids.
