【摘要】 目的 探讨NG-硝基 -L -精氨酸甲酯 (L -NAME)的抗伤害作用及其对异氟醚麻醉作用、大鼠大脑不同脑区突触体一氧化氮合酶 (NOS)活性和一氧化氮 (NO)的影响。方法  30只雄性SD大鼠 ,随机分为生理盐水对照组、L -NAME异氟醚组和L -NAME抗伤害组 ;分别测定给予L -NAME后大鼠的甩尾潜伏期 (TFL)、最大镇痛效应百分率 (%MPE)、异氟醚最低肺泡有效浓度 (MAC)和大鼠不同脑区粗制突触体NOS活性以及NO的生成。结果 L -NAME可延长大鼠的TEL ,增加 %MPE ,降低异氟醚的MAC ,抑制大脑不同脑区突触体NOS活性和NO的生成 (P <0 .0 1)。结论 L -NAME能通过对中枢神经系统突触体NOS活性和NO生成的抑制 ,发挥其抗伤害和减轻异氟醚麻醉作用

【Abstract】 Objective To study the antinociceptive effect of L-NAME and its effect on isoflurane anesthesia by examining the minimum alveolar concentration(MAC) of isoflurane and the activity of nitric oxide synthaes (NOS) and production of nitric oxide(NO) in the synaptosomes of varied parts of rat brain.?Methods Thirty maleSprague-Dawley rats were randomly divided into 3 groups, (1)control group, (2)L-NAMEL-isoflurane group, (3)L-NAME antinociceptive group( n =10 in each group).The tail withdrawal reaction time(TEL), NOS activity and production of NO were determined in groups (1) and (3). The MAC of isoflurane was measured in group (2).? Results L-NAME prolonged TEL, increased %MPE, inhibited the activity of NOS and production of NO as compared with the control ( P <0.01). And L-NAME also decreased rat isoflurane MAC as compared with the baseline ( P <0.01).?Conclusion L-NAME may probably produce its antinociceptive effect and decrease isoflurane MAC by inhibiting NOS activity and NO production in the central nerve system.
