一种分离衰老红细胞的方法:生物素标记法基本原理及其应用

【摘要】 生物素标记法是近年来发展起来的一种分离衰老红细胞的技术。其基本原理是用生物素(NHS-Biotin)标记一定量的红细胞,使之参与血液循环,并在体衰老,利用biotin与抗生物素蛋白(avidin)之间的高亲和力,分离带有标记的、具有特定年龄的红细胞。实验证明这种技术能够精确获得不同特定年龄的红细胞亚群。该技术在临床中可取代51 Cr标记法进行红细胞年龄的测定,并用来进行红细胞在体衰老过程中生物物理、生物化学特性的改变。

【Abstract】 Biotinylation is a new technique which has been developed in recent years to separate se- nescent erythrocytes. Its principle is as follows: red blood cells( RBCs) are labeled with NHS-Biotin, participated in the circulation and ages in vivo. Using the high affinity between biotin and avidin, the labeled RBSs with specific ages are separated. Experiments show that this technique can enable people to precisely get the subpopulation of RBCs with different specific ages. In clinical, it can take place of 51Cr-labeling to measure the RBC’s life span. Furthemore, it can be used to study the biophysical and biochemical changes in the process of RBC’s senescence in vivo.
