随机引物聚合酶链反应用于鉴别嗜人按蚊不同地理株的初步研究

【摘要】 目的探讨嗜人按蚊是否存在亚种(或地理株)。方法应用随机引物聚合酶链反应技术(RP-PCR),选用3种引物对嗜人按蚊江苏株、广西株、广东株的DNA扩增片段进行分析并以中华按蚊江苏株作对照。结果中华按蚊和嗜人按蚊的扩增条带存在明显差别,3种引物对嗜人按蚊江苏株、广西株和广东株的扩增图谱均存在明显差异。结论 RP-PCR显示嗜人按蚊江苏株、广西株和广东株之间存在差异,但这种差异在嗜人按蚊种或种下分类上的意义有待进一步分析。

【Abstract】 Objective To identify the strains of Anopheles anthropophagus. Methods A ran domly primed polymerase chain reaction (RP-PCR) with three selected primers was applied to iden tify Anopheles anthropophagus collected from Jiangsu, Guangdong and Guangxi provinces. The Jiangsu strain of Anopheles sinensis was used as control. Results There is remarkable difference in amplified DNA fragments between Anopheles sinensis and Anopheles anthropophagus. Amplified with three primers, the DNA band patterns of jiangsu, Guangxi and Guangdong strains of Anopheles anthropophagus differed from each other. Conclusion RP-PCR showed the differences among Jiangsu, Guangxi and Guangdong strains of Anopheles anthropophagus. However, the strains classi fication for these differences should be based on further study.
