在体微透析检测胆红素脑病模型鼠EAA神经递质
IN VIVO MEASUREMENT OF THE EAA NEUROTRANSMITTER IN BILIRUBIN INDUCED ENCEPHALOPATHY
【摘要】 探讨胆红素神经毒性脑组织兴奋性氨基酸 (EAA)神经递质变化。应用方法 :制作胆红素脑病动物模型基础上在体脑内微透析、提取神经突触间细胞外液 ,HPLC检测分析天冬氨酸 (Asp)、谷氨酸 (Glu)、甘氨酸 (Gly)。结果显示 :胆红素毒性脑组织细胞外 Gly较对照组明显升高 ,而 Asp、Glu与对照组差异不显著。因此 ,胆红素神经毒性 NMDA受体活性变化机制涉及 Gly细胞外堆积 ,提高 NMDA受体对EAA神经递质 Asp、Glu敏感性
【Abstract】 Study the EAA neurotransmitter concentration in bilirubin induced encephalopathy. Using mthods the basis bilirubin induced encephalopathy in Guinea pig, the Microdialysis Probe was inserted in the hippocmaus zone of the brain and fixed in the Stereotaxic Frame, Kreb’s Ringer was irrigated and extracellular liquid and analysed with HPLC. Results Showed EAA Nurotransmitter GLY was greatly increased in the extracellular liquid of blirubin induced neurotoxicity brain in Guinea pig, RAA Neurtransmitter GLU and ASP not show significant change. EAA Neurotransmitter GLY concentration was increased in the extracellular liquid of the bilirubin encephalopathy brain tissue, thus increased the sensitivity of the ligand ASP and GLU to the NMDA Receptor.
