高氧对缺氧缺血性脑损伤新生大鼠的影响

【摘要】 观察缺氧缺血性脑损伤 (HIBD)新生大鼠长时间吸高浓度氧后脑病理学及DNA损伤的变化。 HIBD新生大鼠吸纯氧 48小时 ,观察脑大体和光镜下的变化 ,并进行脑组织 DNA电泳。结果 HIBD大鼠吸氧后 5天与未吸氧组比 ,左脑丢失重量减少、左脑大体病变程度减轻 ,但均无统计学意义 (P均 >0 .0 5 ) ;吸氧组死亡率高于未吸氧组 ,分别为 11.5 %和 4.2 % ,差异亦无显著性 (P均 >0 .0 5 ) ;HIBD大鼠吸氧后 5天 ,光镜下左脑损伤程度较未吸氧组稍轻 ,但未能进行统计学处理 ;DNA电泳结果表明 ,缺氧缺血后 48小时皮质、海马、纹状体和丘脑的 DNA电泳在模糊涂片带的基础上均可见“梯形条带”,表明既有细胞坏死 ,又有细胞凋亡存在。缺氧缺血后再吸氧 48小时相同脑区的 DNA电泳与未吸氧组相比无明显差别。结论 HIBD新生大鼠吸纯氧 48小时后脑病理及 DNA损伤无明显改善 ,死亡率反而有所增加

【Abstract】 To explore the pathologic and DNA changes in the brain of the hypoxic ischemic brain damage(HIBD)newborn rats after exposed to high oxygen for long time.The pathologic changes were observed by brain general examination and light microscope after HIBD newborn rats were exposed to pure oxygen for 48 hours.The electrophoresis of DNA extracted from brain tissue was used to detect injured DNA Results Shoused that fter HIBD newborn rats were exposed to oxygen,left hemisphere weight and weight dificit,degree of the left cerebral general damage improved slightly on day 5, but there were on differences form on oxygentreated control group.The death rate of oxygen treated group was higher than control group which parameters were 11.5% and 4.2% respectively,but there was on signifincant difference also.Observed by light microscope on day 5 after oxygentreated,the degree of the brain admage was less severe than untreated control group,but statistical analysis could not been made.After newborn rats were hypoxic ischemic for 48 hours,smears and “ladders”of DNA extracted from cortex,hippopcampus,striatum and thalamus appeared on electrophoresis indicating there were sacrified on the same day,the appearance of the electrophoresis of extracted DNA from the brain had no significant changes compared with untreated control group in the same template.Therefose exposure HIBD newborn rats to pure oxygen for 48 hours could not improve brain pathologic and DNAdamage,but the death rate increased.
