水稻叶肉原生质体的分离与培养

【摘要】 研究了秧龄、纤维素酶浓度、纤维素酶与半纤维素酶和果胶酶的比例等因素对分离水稻叶肉原生质体的影响 .建立了培养水稻无菌苗、用叶鞘分离水稻叶肉原生质体的方法 .该方法解决了愈伤组织诱导困难 ,分离水稻原生质体周期长、费时、费力的问题 ,大大缩短了分离原生质体的时间 .对水稻叶肉原生质体的培养条件也进行了初步的研究 .

【Abstract】 The effects of different factors, such as the growing days of the rice seedlings,the concentration of cellulase,hemicellulase and pectolyase Y23,the different mixture of cellulase with hemicellulase and pectolyase Y23 on the protoplastic production, were studied.The results showed that the sheath of the rice seedling growing in 8 to 10 days was the best material for separating mesophyll protoplasts,and the mixture of 2.0% cellcelase,1.0% hemicellulase and 0.5% pectolyase Y23 was most effective enzymatic solution for separating protoplasts from the sheaths of the rice seedlings.The cultural conditions of rice mesophyll protoplasts were also studied.The results showed that the genotype of the rice seedlings was probably one of the main factor which regulates the differentiation of the protoplasts separated from the sheath of the rice seedlings.This protoplast\|separating method resolved the difficulty of inducing Callus and the difficulty that the Callus grow too slowly so that it needs too much time to get a large amount of Callus used to separate protoplasts.In the result,much time to separate protoplasts from the sheth of the rice seedlings can be saved.
