几种基因转移技术在小麦上的应用
Research on gene transformation in wheat
【摘要】 采用基因枪法、电脉冲法、农杆菌介导法和浸泡法将外源基因质粒pZFX2导入小麦细胞中 ,经卡那霉素筛选 ,获得了抗性的愈伤组织和再生植株。通过此项试验 ,研究了不同方法的转化效果 ,摸索了适宜的小麦基因转化条件
【Abstract】 Foreign plasmid pZFX2,containing the NPT Ⅱ(neomycin phosphotransferase)and glycolate oxidase gene which can increase the wheat plant resistance to insects,was introduced into the calli and mature embryos of wheat by the following methods:particle bombardment;electroporation,Agrobacterium mediated transformation and soaking in DNA solution.After selection culture and regeneration culture,some transgenic calli and plants resistant to Kanamycin were obtained.The optimum conditions and the efficiency of these gene transformation methods were studied.
