西番莲胚乳愈伤组织诱导和三倍体植株再生
CALLUS INDUCTION AND TRIPLOID PLANT REGENERATION FROM ENDOSPERM OF PASSIFLORA EDULIS
【摘要】 用紫果西番莲种子的胚乳作外植体离体培养 ,在MS +BA 0 .2mg/L +NAA 2 .0mg/L培养基上诱导获得乳白色或淡黄色结构致密的愈伤组织 ,诱导频率达到 84.7%。愈伤组织在MS +BA 2mg/L +NAA 0 .2mg/L上分化不定芽的频率最高 ,达 6 5 % ,增殖系数达到 5 .2。再生的不定芽在 1/ 2MS+IBA 1mg/L上诱导生根的效果好 ,生根率达 6 3%。再生植株经根尖染色体计数有 95 .3%的细胞染色体数目为 2n =3x =2 7,证明再生植株为三倍体。说明胚乳培养是获得三倍体的高效途径
【Abstract】 Endosperm of Passiflora edulis was used as explant. The callus induction rate (84.7%) was obtained when endosperm was cultured on MS supplemented with 0.2mg/L BA and 2.0 mg/L NAA.The highest shoot induction rate(65%)was abtained when callus was cultured on MS+BA 2.0 mg/L+NAA 0.2 mg/L and multiplication coefficient was 5.2. The shoots rooted readily on 1/2 MS supplemented with 1mg/L IBA and the root rate was 63%.The regenerated plantlets proved to be triploidy(2n=3x=27).
