中国鲎鲎素对人胃腺癌BGC-823细胞形态和超微结构的影响

【摘要】 应用从中国鲎血液中提取的鲎素处理人胃腺癌 BGC- 82 3细胞 ,研究鲎素对胃癌细胞形态和超微结构的影响 .光镜观察显示经 2 .0 μg/m L鲎素处理的 BGC- 82 3细胞形态较为一致 ,细胞体积增大 ,趋于扁平铺展状态 ,细胞核质比例变小 ,核仁数量减少、体积缩小 .扫描电镜与透射电镜观察可见经鲎素处理后 BGC- 82 3细胞表面微绒毛稀少 ,细胞边缘丝状伪足减少 ,片状伪足增多 ;细胞核形态较规则 ,核内异染色质减少 ,常染色质增多 ;细胞质内线粒体数量增多 ,结构较为一致 ,高尔基体呈较为典型发达状态 ,并出现粗糙型内质网增多和多聚核糖体减少等变化 .结果表明 ,鲎素能有效改变胃癌细胞恶性形态结构特征 ,从而对胃癌细胞具有一定的诱导分化作用

【Abstract】 In this study, we used tachyplesin which is isolated from acid extracts of horseshoe crab (Tachypleus tridentatus) hemocytes, to treat the human gastric adenocarcinoma BGC823 cells, and investigated the effects of tachyplesin on the morphology and ultrastructure of BGC823 cells. Optical microscope observation showed that BGC823 cells had undergone restorational morphological changes after being treated with 2.0 μg/mL tachyplesin. The volume of cells increased, the nucleocytoplasmic ratio decreased, the number and volume of nucleolus decreased. Scanning electron microscope and transmission electron microscope observation showed that, in the BGC 823 cells which were treated with 2.0 μg/mL tachyplesin, microvilli and filopodia reduced, sheed pseudopodia increased, the shape of nuclear became regular, heterochromatin decreased while euchromatin increased, the number of mitochondrion increased and its structure appeared consistent, Golgi complex turned to be typical, endoplasmic reticulum increased and polyribosome decreased. These results showed that tachyplesin could change the malignant morphological and structural characteristics of the human gastric carcinoma cells effectively and had inducing differentiation effects on the human gastric carcinoma cells.
