Bt转基因水稻“克螟稻”杂交后代二化螟抗性研究初报

【摘要】 利用室内叶片快速鉴定法结合田间抗性调查 ,研究了粳型 Bt转基因水稻“克螟稻”与籼、粳稻杂交后代对二化螟 ( Chilo suppressalis)的抗性。结果显示 ,在与籼稻和粳稻的杂交后代 ( F1、F2 和 BC1)中 ,都出现了对二化螟表现高抗的植株。用离体叶片法测定 ,4个组合 12 0株杂种 F1和 88株 GUS阳性( GUS+) BC1株与 30株克螟稻亲本相似 ,2天内对二化螟初孵幼虫的死亡率高达 10 0 .0 % ,而用相应组合的 GUS阴性 ( GUS-)株和非转基因亲本叶片饲喂的二化螟初孵幼虫 4天内的死亡率仅为 8.33%~ 2 2 .8%。此外 ,在未施用对鳞翅目害虫有毒性的农药的试验田 ,共调查 8个组合 70 7株 GUS+F2 植株和 7个组合 177株 GUS+BC1植株 ,仅有大约 1%的植株出现枯心和白穗 ;而对应的 4 0 5株 F2 和 182株 BC,GUS-株中分别有 99.3%和 99.5%出现了枯心。本实验的结果表明 ,应用杂交、回交常规育种方法将克螟稻的抗虫基因导入到新的推广水稻品种是可行的 ;同时也反映了报告基因 GUS仍可作为检测 cryl Ab的简便而有效的手段。

【Abstract】 Laboratory bioassay and field evaluation were conducted to investigate the resistance to stripe stem borer (SSB, Chilo suppressalis ) of the progenies of transgenic rice lines KMD1 and KMD2 containing crylAb gene crossed to conventional rice varieties. The results indicated that, there were plants with high resistance to SSB in all progenies of F 1, F 2 and BC 1 from crosses of transgenic rice to both indica and japonica rice. Using cut leaf feeding test, newly hatched larvae died 100.0% within 2 days when fed leaf tissues of 120 F 1 and 88 GUS positive BC 1 plants, while only 8.3% to 22.8% larvae died even by the fourth day fed leaf tissues from GUS negative control plants. In the field without pesticide spray, about 99% of 177 GUS positive BC 1 plants and 707 GUS positive F 2 plants were not damaged by SSB, while 99.3% and 99.5% of GUS negative F 2 (405) and BC 1 (182) plants were found with dead heart(s), respectively. The results indicated that it is possible to introduce transgene crylAb into other commercial varieties through crosses and back crosses, and GUS assay could be a simple method to monitor crylAb gene expression and assist selection for resistant plants.
