芥菜真空渗入遗传转化体系适宜苗态的建成
Forming of Optimum Plant Shape of Mustard for High Efficiency Genetic Transformation System with Vacuum Infiltration Method
【摘要】 通过 0~ 4℃低温处理芥菜种子和春秋 7个播期试验 ,研究了根癌农杆菌菌株 GV31 0 1 (携带 Ca MVCABB-BJI和 Bari-1基因 VI)和 LBA4 40 4 (携带 GUS基因 )转化芥菜的真空渗入高效遗传转化体系的适宜苗态建成方法。芥菜种子经 50℃温烫 1 0 min、2 3℃浸种 2~ 6h、催芽 2 0 h,或 50℃温烫 1 0 min、2 3℃浸种5h、不催芽 ,然后置 0~ 4℃低温下处理 2 5d,种植大田 ,在长日照条件下易形成适宜真空渗入遗传转化苗态 ;圆叶芥 9月 2 0日、9月 30日和次年 2月 2 7日露地播种 ,雪里蕻 9月 30日和次年 2月 2 7日露地播种 ,易形成真空渗入遗传转化的适宜植株形态
【Abstract】 By treating the seeds of mustard ( B. juncea Coss.) at 0~4℃ and sowing in seven different dates in autumn and spring, the ways to form optimum plant shape are studied for high efficiency genetic transformation system with vacuum infiltration method on the GV3101(carrying CaMV CABB BJI and Bari 1 gene VI) and LBA4404 (carrying GUS gene) strain of Agrobacterium tumefaciens . Seeds at 50℃ hot water being warmed for 10 min, at 23℃ soaked for 2~6 h, at 23℃ sprouted for 20 h or at 50℃ hot water being warmed 10 min, at 23℃ soaking 5 h, then at -4℃treated for 25 days, after that planted in soil are easy to form optimum plant shape for vacuum infiltration genetic transformation during long day light. Cv. Yuanyejie seeds being sowed in open field from 20 September to next year on 17 February and cv. Xuelihong, seeds being sowed in open field from 30 September to next years on 17 February are easy to form optimum plant shape for vacuum infiltration genetic transformation.
