子房注射法获得转基因抗虫棉植株研究
Insect resistant Transgenic Cotton Plants Transformed with Pollen Tube Pathway
【摘要】 采用子房注射法将构建在植物表达载体 p BI1 2 1 .1上的 Bt杀虫基因导入棉花品种石远 3 2 1、中植3 72、82 2和辽棉 90 0 1中 ,获得 8株转基因植株。经温室抗虫性鉴定 ,8株转基因植株对棉铃虫都表现出较好的抗性。同时对这 8棵棉株叶片进行的 PCR检测结果证明 Bt基因已经整合进植物基因组中
【Abstract】 By injecting a synthetic Bt crystal protein gene, GFM Cry1A, cloned in a plant expression vector, pBI121.1, into ovaries of cotton varieties Shiyuan321, Zhongzhi 372, 822 and Liaomian9001, 8 Bt transgenic plants were obtained. A bioassay was performed in a greenhouse. The plants showed satisfactory resistance to larvae of bollworm. The positive result of PCR proved that the Bt gene had been integrated into cotton genome. These results indicated that such direct transformation in situ as an ovary injection mathod was efficient.
