人胚胎成纤维细胞的冷冻保存
Cryo-preservation of Human Embryonic Fibroblasts
【摘要】 将 6~ 1 4代的人胚胎成纤维细胞以 0 .5、0 .62、0 .89和 1 .1 2℃ /min的降温速率用胚胎冷冻法进行超低温冷冻保存 ,解冻活率达到 0 .83~ 0 .93。并证明平衡时间以 2~ 2 .5h为宜 ,诱发结晶步骤可以省略。其饲养层寿命长达 2 5~ 2 8d,还可反复冷冻 ,长期应用于胚胎干细胞的分离与克隆研究。
【Abstract】 Human embryonic fibroblasts (hEF) of passage 6~14 were cryo preserved by method of embryo freezing with temperature decreasing rate of 0.5℃,0.62℃,0.89℃ and 1.12℃ per minute. The cell motility thawed was 0.89~0.93.The result suggests that the proper equilibrium duration is 2~2.5 hours, ice seeding is not necessary. The feeders of hEF could last for 25~28 days. It is concluded that thawed hEF could be cryo preserved and utilized in the isolation and cloning research of embryonic stem cells for long time.
