β-胡萝卜素和维生素C对Ni2O3诱导人肺成纤维细胞转化的保护作用
Protection of β-carotene and vitamin C on the transformation of human lung fibroblasts cell induced by nickel sesquioxide
【摘要】 为了探讨三氧化二镍 (Ni2 O3)诱导人肺成纤维 (HL F)细胞恶性转化作用以及β-胡萝卜素和维生素C的保护作用 ,用不同浓度的 Ni2 O3在体外多次处理 HL F细胞 ,利用刀豆凝集素 A(Con A)凝集实验和软琼脂集落形成实验进行转化细胞的恶性鉴定 ;同时添加β-胡萝卜素 (5 .4mg/ L )和维生素 C的食物 (1.8mg/ L ) ,观察对 Ni2 O3转化作用的影响。结果表明 Ni2 O3(0 .5~ 2 .0 mg/ L)可诱导 HL F恶性转化 ,转化细胞生长速度加快 ,排列紊乱 ,失去接触抑制 ,呈交叉重叠生长 ,转化率呈剂量 -效应关系。转化细胞可被低浓度 Con A凝集 ,并在软琼脂内生长。添加 β-胡萝卜素和维生素 C的处理组比单用 Ni2 O3 组的转化率显著下降 (P<0 .0 5 ) ,处理后细胞 Con A凝集反应阴性 ,不能在软琼脂内生长。结论认为 Ni2 O3有较强的诱导 HL F细胞恶性转化的能力 ,具有致癌性。β-胡萝卜素和维生素 C对于镍诱导 HL F恶性转化具有保护作用 ,提示职业接触镍的人群有必要在膳食中增加富含β-胡萝卜素和维生素 C,以提高机体的抗氧化能力 ,从而提高机体的抗肿瘤能力
【Abstract】 In order to study the protection ofβ- carotene and vitamin C against the transformation of human lung fibroblasts(HL F) cell induced by nickel sesquioxide,HL F cells were treated repeatedly by different concentrations of Ni2 O3in vitro.β- carotene(5 .4m g/L) and vitam in C(1.8mg/L) were added into the media containing Ni2 O3(1.0 mg/L ) respectively.The identification of malignancy of the transformation of HL F cell was carried out by the tests of Con A and the growth on semisolid agar culture.The results showed that Ni2 O3could induce the m alignanttransform ation of HL F cell.The transformed cell proliferated rapidly.The transformed colonies exhibited in extensively random orientation and the cells were crossing- over.The frequency of transform ation showed a dose- response relation at the experim ental concentrations. The transformed cell could be agglutinated by lower concentration of Con A and could grow in semisolid agar.The frequencies of transformation of HL F cell exposed toβ- carotene and vitamin C were decreased significantly.The cells could not be agglutinated by Con A and notgrow in sem isolid agar.It was concluded that Ni2 O3 could induce strongly the malignant transformation of HL F cell and might be carcinogenic to human.The protection ofβ- carotene and vitamin C on the transformation of HL F cell induced by Ni2 O3was observed.The increase of foods riched inβ- carotene and vitamin C was suggested for workers exposed to nickel.
