镉的免疫毒性与促肾上腺皮质激素释放因子的关系
Study on the relationship between cadmium immunotoxicity and corticotropin releasing factor
【摘要】 对 Cd Cl2 和 Cd Cl2 + α- h CRF(α螺旋促肾上腺皮质激素释放因子 )分别染毒的 SD大鼠 ,用 [3H-Td R]掺入法测定大鼠脾 T、B淋巴细胞增殖转化功能 ,垂体前叶细胞培养法测定中枢下丘脑和外周血浆中的 CRF含量。结果表明单独染镉组有丝分裂原诱导的鼠脾 T、B淋巴细胞增殖转化均受到明显抑制 ,具有剂量 -效应关系 (P<0 .0 5 ) ;动物下丘脑 CRF含量伴随着染镉剂量升高 (P<0 .0 5 ) ;外周血浆 CRF含量各组间差异无显著性 (P>0 .0 5 )。当使用拮抗剂 α- h CRF后 ,在含镉染毒各剂量组间鼠脾 T、B细胞功能抑制差异无显著性 (P>0 .0 5 ) ;下丘脑 CRF含量在各含镉联合染毒组间差异无显著性 (P>0 .0 5 ) ,但均较对照组高 (P<0 .0 5 ) ;外周血浆 CRF水平有升高趋势。结果提示单独染镉可引起下丘脑 CRF含量升高和免疫功能抑制 ,联合使用 α- h CRF后则能部分改善镉对大鼠脾细胞的免疫功能抑制。镉对大鼠脾淋巴细胞免疫毒作用过程中存在中枢 CRF介导的免疫调节机制
【Abstract】 To explore the relationship between cadmium immunotoxicity and corticotropin releasing factor(CRF), murine T and B lymphocytes mitogen induced proliferation function( 3H TdR incorporation method) and CRF levels of hypothalami and blood plasma were observed both in cadmium chloride(CdCl 2) and CdCl 2+α hCRF treated animals in vivo. In cadmium treated animals, T and B limphocytes stimulation indexes were significantly lower than those in control( P <0 05) with a dose dependent response. And interestingly, the central CRF levels in CdCl 2 treated groups were significantly higher than those in control( P <0 05), an increasing CRF level was also observed accompanied by the increase of CdCl 2 level. In peripheral blood plasma, no significant difference of CRF levels was found among all groups( P >0 05). When animals were treated with CdCl 2 in combination with α hCRF, no significant difference of B lymphocyte proliferation function existed among the α hCRF+CdCl 2 groups ( P >0 05). Similar results were also observed in detecting T lymphocyte proliferation function. Hypothalamic CRF levels among various doses of CdCl 2+α hCRF groups showed no statistical difference, but all were higher than those in the control, and peripheral CRF levels had a trend to increase in α hCRF+CdCl 2 co administrated groups. The results suggested that CdCl 2 showed an immunosuppressive action upon murine T and B lymphocytes, CdCl 2 could cause central CRF increase. α hCRF could partially reduce the cadmium immunotoxicity. Central CRF mediated immunomodulation was shown in cadmium immunotoxical effects on murine splenic lymphocytes.
