选育糖化酶产生菌简捷方法的研究

【摘要】 糖化酶是应用历史悠久用途广泛的酶类,但其产生菌的传统筛选方法工艺繁荣,而且无法排除α-淀粉酶和葡萄苷转移酶的影响,筛选效率低。本文报导一项筛选糖化酶产生首快速方法的国家发明专利(专利证书号13016)的研究过程。其特点是选育到1株能反映糖化酶产量的指示菌Ay菌株(candidakrusei),它可在平板上检测大量持选菌的单菌落琼脂柱,同时又能在α-淀粉酶和葡糖苷转移酶同时存在时,排除它们的干扰,依糖化酶产生菌筛选效率大大提高。该专利技术方法简单,设备易找,投资少,不但对菌种选育单位有意义,而且对酶制剂工厂常规育种和发酵条件的优选都是很有价值的。

【Abstract】 Glucoamylase has long been utilized extensivey. However, the traditional breeding method of glucoamylase producing strains is complicated, and the effect of α-amylase and glucosidetransferase can not be ruled out. A new tapid breeding method is discussed in this paper. In addition, this new method has been got a National Invention Patent for (Pat. no. 13016). The characteristic of the method is that an indicator strain of Ay (Candida krusei) which can reflect the output of glucoamylase has been bred. The indicator strain can be used to check out a large number of under-screening singe colonies agar coumn on agar plate.Moreover, it can rule out the interference of α-amylase and glucocidetransfrase under the circaretance of their coesistence. Therefore, it can raise the efficiency of screening process. It is simple, uses easy-to-get equipment and has low investment. It is not only good for strain screening unit but also valuable for enzyme producing factory in regular strain breeding and fermentation conditions.
