茶多酚对矽尘处理大鼠肺胶原mRNA表达水平的影响
The effect of green tea polyphenols on expression levels of pulmonary collagen mRNA in rats treated by silica
【摘要】 【目的】 从分子水平探讨茶多酚 (GTP)抑制矽肺纤维化的机制。【方法】 使用人Proα1(Ⅰ )、Proα1(Ⅲ )型胶原cDNA探针 ,采用Northern杂交技术 ,观察矽尘处理大鼠在饮用不同剂量茶多酚饮水后 ,肺胶原mRNA水平。【结果】 经各剂量茶多酚处理染尘大鼠Proα1(Ⅰ )和Proα1(Ⅲ )表达水平均不同程度低于矽肺对照组大鼠 ,14mg/mlGTP组下降最为明显。【结论】 GTP可能抑制矽肺肺胶原基因表达。
【Abstract】 Objective To Study the inlibition effect of GTP on the molcular mechanism of fibrogenesis in silicosis. Methods \ Human Proα 1(Ⅰ) and Proα 1(Ⅲ) cDNA probes and Northern Blot hybridization were employed to determine the lung collagen mRNA levels in silica treated rats, given various doses of green tea polyphenols(GTP) in drinking water. Results \ It showed that Proα 1(Ⅰ) and Pro α 1(Ⅲ) mRNA levels of silica\|treated rats given various doses of GTP were lower,particularly at dose of 14 mg/ml GTP,than those of the silicosis rats. Conclusion \ It was saggested that GTP may inhibit the gene expression of lung collagen in silicosis.
