玉米×二倍体多年生类玉米后代的GISH分析

【摘要】 通过对玉米自交系旅 9与二倍体多年生类玉米 ( Zea diploperennis,DP)远缘杂交 ,其杂种与玉米回交 ,回交一代 ( BC1 )孤雌生殖 ,获得了具有 DP性状的两个稳定株系 a2 - 1和 a2 - 6 .对其中的外源渗入片段进行了荧光原位杂交物理定位 ,确定了 a2 - 1中外源渗入片段在第 1染色体长臂近末端 ,第 2 ,8染色体长臂中部 ,以及第 6染色体短臂中部 .a2 - 6中外源渗入片段在第 1染色体长臂近末端 ,第 2 ,8染色体长臂中部 .对两株系中渗入片段在染色体上的物理位置的特点 ,位置与抗病性之间的关系 ,以及可能携带有抗病基因的外源渗入片段等进行了初步的分析和讨论

【Abstract】 After crossing maize inbred line Lu 9 with Zea diploperennis and backcrossing their F 1 with Lu 9, the BC 1 was reproduced with parthenogenesis and two stable alloplasmic pure lines a 2\|① and a 2\|⑥ were obtained. We have physically located the introgressed segments of DP in lines a 2\|① and a 2\|⑥ by genomic in situ hybridization(GISH). In line a 2\|①, the introgressed segments were located on the subterminal region of the long arm of chromosome 1, middle parts of the long arms of chromosomes 2 and 8, and middle part of the short arm of chromosome 6, and in line a 2\|⑥, they were located on the subterminal region of the long arm of chromosome 1, middle parts of the long arms of chromosomes 2 and 8. The characteristics of the physical locations for DP introgressed segments in maize genome, the relation between their distribution on the chromosomes and behavior of different disease resistances, and a giving introgressed segment probably containing which kind of disease resistant gene have been discussed.
