应用RT-PCR技术检测人类甲状腺癌中ret癌基因活化的研究

【摘要】 目的 观察人类甲状腺癌中ret癌基因的活化及其意义。方法 应用RT PCR技术研究甲状腺癌新鲜组织中ret癌基因活化形式PTC基因的表达。结果  2 5例甲状腺乳头状癌中有 6例 (2 4% )PTC基因表达阳性 ,主要分布于Ⅱ级以上肿瘤中 ;而甲状腺滤泡型癌、甲状腺腺瘤和正常甲状腺组织中PTC均为阴性。结论 PTC基因是ret癌基因新的活化形式 ,ret基因的活化仅限于甲状腺乳头状癌类型中 ,可能是乳头状癌的特殊遗传事件 ,因此检测PTC基因可作为人类甲状腺乳头状癌鉴别诊断和判断其生物学行为的有效参考指标

【Abstract】 Objective: To investigate the frequency and significance of ret oncogene activation in human thyroid carcinomas. Methods: Expressions of PTC oncogene which is a novel rearanged version of the ret tyrosine kinase proto oncogene were detected in fresh tissues of human thyroid carcinomas by RT PCR. Results: We found the presence of an activated form of the ret oncogene in 6 (24%) of 25 papillary carcinomas. By contrast, none of follicular carcinomas, benige lesions and nomorl tissues of thyroid showed PTC positive. Conclusion:PTC oncogene is a novel activated form of ret oncogene and restricted to the papillary cancer subtype. From this findings, we would suggest that ret activation may reprsesnt a specific genetic event that may be of value in the differential diagnosis and determination of biologic behaviour of human papillary carcinomas.
