栝楼毛状根培养与天花粉蛋白合成的研究
CULTURE OF TRICHOSANTHES KIRILOWII HAIRY ROOTS AND TRICHOSANTHIN BIOSYNTHESIS
【摘要】 通过 L BA94 0 2农杆菌转化 ,诱导出了栝楼的毛状根 ;经过高压纸电泳检测甘露碱表明 ,农杆菌 Ri质粒上的 T- DNA已经进入栝楼细胞 ,并且表达合成了甘露碱 ;经实验选出了生长速度较快的 A8转化根系 ;MS培养基 ,2 5℃培养温度 ,350 lx弱光条件有利于毛状根的生物量积累 ,新鲜毛状根的天花粉蛋白含量为 0 .5~ 0 .6mg/ g。光照条件不利于天花粉蛋白的合成。
【Abstract】 The formation of hairy roots of Trichosantheskirilowii Maxim.was induced by the transformation of Agrobacterium rhizogenes LBA940 2 .The T- DNA( transfer DNA) on the Ri plasmid was affirmed to transfer into the cell of T.kirilowii by the high- voltage paper electrophoresis which could detect the mannopine’s existence.The best hairy roots- A8was selected and cultivated on the MS medium,2 5℃ and35 0 lx light intensity,which were the optimum condition.On thatcondition,A8grew very well and the trichosanthin concentration of the fresh hairy roots was also comparatively high- about 0 .5~ 0 .6 mg/g,although Trichosanthin biosynthesis was inhibited in the light.
