甾体11β-羟基生物转化新工艺的研究
STUDY ON BIOCONVERSION TECHNOLOGY OF 11β-HYDROXYLATION OF STEROIDS
【摘要】 通过分析 11β-羟基转化体系中反应介质和发酵液的影响 ,设计了一种将化合物 RS- 2 1醋酸酯转化为氢化可的松的新工艺。确定工艺条件为 :孢子接种浓度 1× 10 6/ m L ,2 8℃ ,摇床转速 (150~ 170 ) r/ min,培养 (17~ 2 1) h,培养液 p H值降至 3.8、菌体处于稳定期前期、菌丝干重达 7.2 mg/m L以上时 ,以 1∶ 1稀释比稀释 ,投料转化 2 4 h,β体转化率达 (75~ 76 ) %。初步研究了新工艺的转化过程特性
【Abstract】 WT5”BZ]According to effect of the reaction medium and cultural medium on 11β hydroxylation,the new bioconversion technology was established which Reichsterins substance acetate was converted to hydrocortisone by 11β hydroxylation.The effect facts were observed.The process conditions were:sports concentration 1×10 6/mL,temperature 28 ℃,rotation speed of shake flask (150~170) r/min,hypha age (17~21) h,pH volute of culture fluid 3.8,hypha weight (dry weight) 7.2 mg/mL,rate of dilution 1∶1,fermentation periods 24 h.The rate of conversion of hydrocortisone was 75%~76%.The 11β hydroxylation character of new technology was studied.
