大承气汤颗粒剂对肠源性内毒素血症大鼠重要脏器一氧化氮合酶的影响

【摘要】 目的 :研究大承气汤颗粒剂 (简称颗粒剂 )对肠源性内毒素血症大鼠肝、肠、肠粘膜组织中一氧化氮合酶 (NOS)活性的影响。方法 :大鼠腹腔注射大肠杆菌悬液,造成腹膜炎诱发的肠源性内毒素血症模型 ,分光光度法测定大鼠肝、肠、肠粘膜组织中的NOS活性。结果 :肠源性内毒素血症大鼠 ,肝、肠、肠粘膜组织中NOS活性明显高于对照组 (P<0.05) ;不同剂量的颗粒剂均能显著降低内毒素血症所致的NOS活性升高 ;头孢克洛 (125mg/kg)无效。结论 :颗粒剂能降低内毒素血症时重要器官组织中的NOS活性 ,这可能是大承气汤防治中毒性休克 ,乃至多脏器衰竭的重要生化机理之一。

【Abstract】 Objective:To study the activities of nitric oxide synthase(NOS)in the liver、intestine and intestinal mucosa under experimental enterogenous endotoxemia(EE)in rats. Methods:Escherichia coli suspensions were administered ip. in order to induce the peritonitis-related EE and the activities of NOS were measured by spectrophotometry. Results: The activities of NOS in liver、intestine and intestinal mucosa were significantly increased in EE group,conmpared with control group(P<0.05);Varied doses of Da-Cheng-Qi-Tang(DCQT)pellet could alleviate the increased activities of NOS under EE condition,but no effect was seen in cefaclor-treated (125mg/Kg)group. Conclusion:DCQT pellet can decrease the activities of NOS of the important organs in EE induced by Escherichia coli infection. It is one of the biochemical mechanisms of the treatment of endotoxic shock,even multiple organ failure.
