【摘要】 目的　建立人前列腺癌裸鼠自发转移模型 ,观察其生长和转移的生物学特性。方法　利用外科原位移植的技术 ,取材于人前列腺癌细胞系PC - 3M裸鼠皮下移植瘤 ,保持肿瘤组织结构的完整性 ,通过显微外科手术植于裸鼠前列腺两腹侧叶之间 ,濒死时处死。结果　动物平均 30d衰竭 ,处死 ,原位移植成功率10 0 % ,主动脉旁淋巴结转移发生率 10 0 % ,较PC - 3M肿瘤细胞原位注射及PC - 3外科原位移植主动脉旁淋巴结转移发生率更高。结论　肿瘤细胞的本身转移能力及肿瘤基质共同决定肿瘤转移的发生频率。更多还原

【Abstract】 Objective Establishment of an animal model of human prostate cancer demonstrating spontaneous meastasis by surgical orthotopic implantation to understand the disease and to formulate treatment strategy.Methods We implant histologically intact tumor tissues orthotopically in immunodeficient mice.In this study intact tissue of the human prostate cancer cell line PC-3M,harvested from a subcutaneous tumor in a nude mouse,was implanted to the ventral lateral lobes of the prostate gland in a serise of nude mice.Mice were sacrificed when found moribund,and autopsy and histology were performed subsequently.Results In contrast to orthotopic injectioin of PC-3M and surgical orthotopic implantation of PC-3,surgical orthotopic implantation of PC-3M can cause higher frequency of periaortic lymph node metastasis noted upon histological examination.Conclusion Stromal tissue as well as metastatic cells played a critical role in tumor growth an progression.
