子宫内膜整合素蛋白β3亚基在妊娠中的变化
Changes of Endometrial β3 Integrin during Early Pregnancy
【摘要】 为探讨整合素蛋白 β3亚基在胚胎着床期的作用 ,本实验应用特异性抗整合素蛋白 β3抗体 ,采用免疫和 Western印迹法 ,二氨基联苯胺显色 ,VDS扫描等技术 ,检测妊娠 d3、5、7、1 5小鼠子宫内膜及人分泌期子宫内膜和妊娠早期中整合素β3亚基的表达量。结果 :小鼠子宫内膜整合素蛋白 β3亚基表达量在妊娠 d5明显上升 ,至 d7时达高峰 (与对照组比较 ,P<0 .0 1 )。此后下降 ,d1 5时 ,整合素蛋白 β3的表达量与对照组相比 ,无明显差异。人蜕膜组织和分泌期子宫内膜均有整合素蛋白β3的表达。蜕膜整合素蛋白β3的表达量与胚囊大小无关 ,但比分泌期子宫内膜高 4~ 5倍 (P<0 .0 0 0 1 )。这些结果提示 :整合蛋白 β3亚基在子宫内膜中的特异性变化 ,可能启动、促进胚胎的粘附和着床 ,在着床过程中起重要作用。
【Abstract】 WT5BZ] The objective of this study is to investigate the expression of β 3 integrin during implantation. By using Western blot and immunoblot followed by DAB staining methods, combined with auto densitometric scaning,β 3 integrin was determined with purified antibody against the β 3 integrin both in mouse endometrium on day of 3,5,7,15 and in human decidual tissue at the time when embryonic sac was 6~11 mm,12~17 mm,18~24 mm in diameter during the early pregnancy. Results: The expression of β 3 integrin in mouse endometrium increased obviously on day 5 of pregnancy, and reaches a maximum at day 7, then returned to the control level at day 15. β 3 integrin presented on human endometrium in midsecretory phase and decidua the early pregnancy. Statistic analysis indicated that diameter of embryonic sac and the expression of β 3 integrin were negative correlated. Content of β 3 integrin in decidua during pregnancy was 4~5 times higher than that in the control group ( P <0.0001). Conclusion: The increased expression of β 3 integrin suggests they play important roles in the process of implantation.
