干扰素和腺病毒3型诱导MxA蛋白产生的研究

【摘要】 目的 研究干扰素 (IFN)、腺病毒 3型 (Ad3)对MxA蛋白的诱导作用。方法 采用Westernblot检测WI 38细胞在不同浓度、不同时间IFN诱导下胞浆内MxA蛋白的含量 ;采用流式细胞仪检测不同浓度的Ad3诱导健康人外周血单个核细胞 (PBMC)胞浆MxA蛋白的含量。结果 不同浓度的IFN诱导 6h后 ,WI 38细胞胞浆内均有MxA蛋白的产生 ;不同浓度的Ad3诱导PBMC产生的MxA蛋白均显著高于对照组。结论 IFN、Ad3体外均可诱导MxA蛋白的产生

【Abstract】 Objective To evaluate whether interferon(IFN), adenovirus type 3(Ad3) can induce MxA protein. Methods The content of MxA protein in cytoplasm of WI38 cells treated by IFN for varying doses and time was detected with western blotting; the peripheral blood mononuclear cells (PBMC) were isolated from the healthy people and treated with various doses of Ad3 in vitro and the levels of MxA protein were analysed by flow cytometry. Results Varying doses of IFN can induce MxA protein in WI38 cytoplasm after 6 hours of induction. The level of MxA protein in all groups of PBMC treated by varying doses of Ad3 was higher than that of controls. Conclusion MxA protein can be induced by IFN as well as in PBMC in vitro by Ad3.
