核桃油毒性实验研究

【摘要】 目的 :观测核桃油急性毒性及长期毒性。方法 :急性毒性采用 bliss法测定核桃油的 L D5 0 。长期毒性选用 6周~ 8周健康 Wistar大鼠 6 0只 ,雌雄各半 ,随机分为 3组 :核桃油低剂量组、高剂量组 ,分别以 13g/ kg及 4.3g/ kg灌胃。每天一次 ,对照组以 1m L / kg生理盐水灌胃 ,各组均每天给药一次 ,连续 12周。每天均需观察动物一般行为活动、进食量 ,并在给药期间每隔 2周观察动物体重变化。最后一次给药后 2 4h,将每组 3/ 4的动物麻醉 ,从股动脉采血 ,从膀胱取尿 ,检查血、尿常规 ;肝、肾功能 ;采集心、肝、脾、肺、肾等脏器 ,肉眼观察其病理改变 ,制成病理切片做进一步观察。剩余动物再连续喂养 14d,以备进一步检查。结果 :核桃油小鼠一次灌胃给药的最大耐受量大于 40 g/ kg;核桃油小鼠一次腹腔注射给药的最大耐受量大于 40 g/ kg。与对照组相比 ,两种剂量的核桃油连续给药 12周 ,动物的体重、行为、血、尿常规、肝、肾功能无显著性差别 (P>0 .0 5 ) ;心、肝、脾、肺、肾等脏器也无明显病理变化。结论 :以上结果说明核桃油安全无毒、可靠。

【Abstract】 Objective:To investigate acute and long-time toxicity of walnut oil.Method:Bliss law was introduced to determine LD 50 of walnut oil.60 6~8 weeks Wistar rats with each one half of male and female were selected.The animals were divided into 3 groups:walnut oil 13g/kg,4.3g/kg group,ig and control group,which was administered NS,ig,one time a day for 12 weeks.The behavior,food intake and avoirdupois of animals were observed.24 hours later after drug administration 3/4 of animals were anaesthetised,blood and urine samples were collected to investigate blood and urine routine;liver and renal function.Heart,liver,spleen,lung and kidney were collected to investigate their histopathology.Remaining animals were raised for 2 weeks for further examination.Result:The biggest sufferance dose of walnut oil was beyond 40g/kg.Continuous administration of walnut oil for 12 weeks had no significant effects on blood routine,urine routine;liver,renal function and organic histology.Conclusion:walnut oil is safety and innocuity.
