葡萄感霜霉病基因RAPD标记的序列分析

【摘要】 利用WizardDNAclean -upsystem纯化葡萄感霜霉病基因RAPD遗传标记的DNA片段 ,用细菌质粒 pGEMT -easyvector克隆该片段 ,采用自动荧光DNA测序仪对该片段的核苷酸组成进行双向测序。来自欧洲葡萄粉红玫瑰的葡萄感霜霉病基因RAPD标记由 835对核苷酸及其特定序列组成。所获的感霜霉病基因RAPD标记可以作为合成探针的基础 ,用于葡萄抗病育种过程中的早期选择及品种对霜霉病感病性的鉴定。

【Abstract】 This paper presents the sequence of RAPD marker linked to the downy mildew-susceptible genes in grapes.In grapes.The isolation DNA was purified by Wizard DNA cleanup System and inserted into plasmid pGEM T-vector easy.The sequencing was performed based upon automated fluorescent DNA sequencer(Model ABI377).The RAPD marker from the susceptible cultivar Muscat Rose (Vitis vinifera)consists of 835bp,and it’s restriction-sites were analyzed using PC gene software.It is suggested that the sequence of OPO10-835 might be used as basis for synthesixing the specific PCR primers and the probe for detecting grape downy mildew-susceptibility in molecular assisted selection.
