生物素结合对紫膜结构和功能的影响
THE MODIFICATION EFFECT OF BIOTIN ON STRUCTURE AND FUNCTION OF PURPLE MEMBRANE
【摘要】 为了提高细菌视紫红质(BR)的可组装性 ,使之应用于生物传感器等生物器件 ,利用生物素对紫膜进行修饰 ,使之可以被亲和素识别 ,从而可以定向的固定于固体支撑物表面。实验结果表明 :生物素可以修饰紫膜表面的赖氨酸 ,修饰的程度依修饰的时间不同而有所不同。但即使被修饰24小时的紫膜 ,其表面的赖氨酸仍然没有被完全修饰。同时 ,生物素的修饰不会影响紫膜的结构和功能 ,但是不同的结合位点对M412 的衰减会产生不同的影响。这项研究工作为将来紫膜的组装和应用打下了基础
【Abstract】 In order to improve the operability of bacteriorhodopsin (BR) and make use of it on biological devices, we modified the purple membrane with biotin, so that the purple membrane can be recognized by streptavidin and immobilized on the solid wafer. The results showed that the purple membrane can be modified by biotin and the modification extent was dependent on the modification time. The purple membrane can not be entirely modified by biotin even if the modification time was extended to 24 hours. At the same time, we found that the modification had no impact on the conformation and function of purple membrane. But different sites had different effects on M412. This work might lay the foundation for assembling and application of purple membrane in the future.
