RAPD在蔬菜种子纯度鉴定中应用的反应条件优化
The protocol optimum of RAPD used on the testing of vegetable seed purity
【摘要】 随着分子生物学的发展 ,RAPD(随机扩增多态性DNA)在越来越多的领域得到应用 ,但在蔬菜种子纯度鉴定方面 ,国内外报道较少 .为了更好地将RAPD方法应用于蔬菜种子纯度鉴定 ,就反应条件进行了探索 .以几种蔬菜种子为材料 ,使用进口PCR仪 ,在对几种蔬菜种子进行RAPD分析过程中 ,从DNA提取方法 ,模板DNA量 ,Mg2 +浓度 ,Taq DNA聚合酶的量 ,dNTPs浓度 ,随机引物浓度及反应缓冲液的量等几个方面进行研究 .从而确定了最适的反应条件 ,为RAPD在蔬菜种子纯度鉴定中应用奠定了基础
【Abstract】 With the development of molecular biology,RAPD (Random amplified polymorphic DNA)was widely used in more and more biological fields,but there were fewer reports of using RAPD in the testing of vegetable sees purity in nation and abroad.In order to use RAPD in testing the vegetable seed purity measurement more freely,this paper studied the protocol optimum of RAPD.With the material of several vegetable seeds,the DNA extraction method,template DNA amount,Mg 2+ concentration,Taq DNA polymerase amount,dNTPs concentration,random primers concentration,and PCR reaction files were studied by using PE2400.The best protocol optimum of RAPD used in the testing of vegetable seed purity was decided.
