细胞间粘附分子-1在人胃癌裸鼠移植瘤中的表达

【摘要】 目的 探讨胃癌细胞产生的细胞间粘附分子 1(ICAM 1)在胃癌侵袭和转移中的作用。方法 利用层粘蛋白的粘附性 ,筛选出对Laminin高粘附力 (Lm +)和低粘附力 (Lm - )的两种不同胃癌细胞MKN45亚株 ,并接种于裸鼠体内 ,以免疫组化法观察裸鼠移植瘤组织中ICAM 1的表达。结果 Lm +胃癌细胞株瘤块与周围组织粘连较Lm -瘤块明显。ICAM 1在正常胃上皮无表达 ,瘤块组织有不同程度表达 ,Lm +瘤块ICAM 1表达高于Lm -瘤块 ,阳性染色细胞主要位于瘤块边缘或瘤块离散边缘。结论 胃癌细胞ICAM 1的表达与肿瘤的转移有关

【Abstract】 Objective To appraise the expression of ICAM1 in gastric carcinoma of nude mice and its relationship to invasion and metastasis. Methods Two subpopulations from human gastric cancer cell line MKN45 i.e. lamininadherent cells (Lm+) and lamininnonadherent cells (Lm-) were separated by lamininadhesion method in vitro, and inoculated in nude mice. ICAM1 expression in metastatic tissue of nude mice was detected immunohistochemically. Results Lm+ cancer cell mass showed higher adhesive capacity to surrounding tissue than Lm- ones. ICAM1 expression was scarcely seen in most normal gastric epithelial cells, and was significantly stronger in Lm+ than in Lm- cancer cell mare. Positively stained cells were mainly localized at the edge of the tumor.Conclusion These results implicates the synthesis of ICAM1 by the gastric cancer cells and correlates well with increased risk of tumor invasion and metastasis.(Shanghai Med J, 2000,23∶420422)
