茶多酚对原代培养的大鼠皮肤角朊细胞和成纤维细胞生长的影响

【摘要】 目的 探讨茶多酚对原代培养的皮肤角朊细胞和成纤维细胞生长作用的影响。方法 利用原代培养的两种主要的皮肤细胞———角朊细胞和成纤维细胞,采用丙酮酸比色法、TBA比色法和DTNB(双硫代对硝基苯甲酸)法测定细胞培养的上清液胞浆酶(LDH)释放情况、脂质过氧化产物(MDA)、谷胱甘肽过氧化物酶(GSHPx),并测定培养细胞的生长情况,对茶多酚在皮肤中可能起到的作用进行评价。结果 在加入茶多酚后不同时间,发现茶多酚具有保护细胞膜的功能,即减少LDH的释放、减少MDA产生、增加GSHPx含量、促进细胞生长。结论 体外实验表明茶多酚具有促进皮肤细胞生长、抗脂质过氧化的功效,其最低有效作用浓度为0.05%~0.1%。

【Abstract】 Purpose To observe the possible effects of tea polyphenols on skin cell growth in rat primary cultured keratinocytes and fibroblasts. Methods Pyruvic acid spectrometry,TBA (thiobarituric acid) method,dithiobis p nitrobenzoic acid(DTNB) method and cytometry were used to determine the release of cell plasma enzyme (LDH),lipid peroxidation products (MDA),GSH?Px and cell growth. Results Tea polyphenols can stabilize the membrane of skin cells in concentration from 0.05% to 0.1%,showing in decrease of cell plasma enzyme LDH,MDA,and increase of GSH?Px and cell numbers,significantly. Conclusions It shows that tea polyphenols can improve skin cell growth in vitro and have effective function as anti peroxidation,its lowest proper concentration for cell growth is from 0.05% to 0.1%.
