弓形虫RH、B36、Fukaya株毒力及致病性的比较研究
Comparative Studies on Virulence and Pathogenicity in RH,B36 and Fukaya Strains of Toxoplasma gondii
【摘要】 目的探讨不同毒力的弓形由株与其致病性的关系、方法应用体外培养和动物模型观察RH、B36、Fukaya三虫株对组织细胞的损害及其致病、致畸、致死的差异。结果Vero细胞在8h内的侵袭率及分解产物脂肪酸含量以B36株为高。胞内24h繁殖数以RH株为最多,B36次之,Fukaya最低。此结果与虫体对细胞的损害程度、对仔鼠智力发育的影响密切相关。细胞因子激活巨噬细胞后,对弓形由抑虫或杀虫程度与NO浓度相一致。结论RH株NO浓度高,胞内虫株可被完全消灭,NO可能参与抗虫作用。
【Abstract】 Objective The comparison on virulence and pathogenicity in RH,B36 and Fukaya strain of Toxoplasma gondii was investigated. Methods By using in vitro cultivation and animal model, differences of the 3 strains, RH, B36, Fukaya, in causing tissue and cell damage, motality, criteria of physical and mental pathogenesis as well as frequency of fetal anomalies were examined. Results The parasitic invasibility to cultivated Vero-cells at 8h after infection was found the highest in B36, in which, a parallelly increased metabolic product content of fatty acids was demonstrated, the number of proliferated parasites in the host cells within 24h appeared to be the most in strain RH with a moderate in B,, and the lowest in Fukaya. The result showed a positive correlation with the degree of cell damage and the influence of intelligence in the off-springs. The intensity of parasitocidal and inhibition effect of cytokine activated M against Toxoplasma was shown to be concomitently associated with the reactive NO concentration detected in the medium. Conclusions Along with a complete elimination of the parasites in the host cells, the RH strain gave a higher NO concentration than the other 2 strains, which indicats that NO plays a role in the anti-parasite mechanism.
