【摘要】 目的 :探讨喉癌基因 c- erb B- 2点突变及过表达同喉癌发生发展的关系。方法 :应用 PCR- SSCP对c- erb B- 2的点突变进行检测 ,采用免疫组化法分析 c- erb B- 2的过表达。结果 :40例喉癌阳性表达率为 52 .5%( 2 1 / 4 0 )。其中 2 4例声门上癌表达率为 57% ( 1 7/ 30 ) ;6例声带癌表达率为 50 % ( 3/ 6) ,4例声门下癌表达率为 2 2 .5% ( 1 / 4 ) ,同前者差异有高度显著性 ( P<0 .0 1 ) ,未见 c- erb B- 2的点突变检出。结论 :喉癌中广泛存在着 c- erb B- 2的扩增及过表达 ,而喉癌细胞中却无 c- erb B- 2点突变 ,推测可能存在着另外的基因改变或 c-erb B- 2的扩增和过表达为喉癌发生的主要原因。更多还原

【Abstract】 Objective: To investigate the overexpression and point mutation of c erbB 2 involved in the carcinogenesis process of laryngeal squamous carcinomas.Methods: Using laryngeal squamous carcinoma specimens from 40 patients.The point mutation of c erbB 2 was determined by PCR-SSCP;the over expression of c erbB 2 was analyzed by immunohistochemical methods.Results:Of 40 laryngeal carcinoma specimens,the overexpression of c erbB 2 were detected in 18 cases,the positive staining was 52.5%(21/40);the positive staining of 21 supraglottic carcinomas and of 6 glottic carcinoma were 57% (17/30) and 50%(3/6) respectively,while the positive staining of 4 subglottic carcinoma was only 22.5%(1/4),the difference was significant (P<0.01).In comparison with the former,no point mutation was observed in these specimens.Conclusion:There are extensive overexpression and amplification of c erbB 2 in laryngeal carcinoma,but no detection of point mutation.This suggests that other kind of activation probably has not been found,or the over expression and amplification of c erbB 2 might be the main reason that promotes laryngeal carcinoma.更多还原
