大鼠血清中二十碳五烯酸与二十二碳六烯酸的气相色谱测定法研究

【摘要】 目的 :用气相色谱测定法观察改变大鼠饲料成分后 ,其血清中二十碳五烯酸 (EPA)与二十二碳六烯酸 (DHA)组分含量的变化。方法 :选用丁二酸二乙二醇酯 (DEGS)色谱柱。样品经提取后用三氟化硼 -乙醚 /甲醇 ,甲酯化处理。结果 :两者酯化率均为 97.1%~ 10 0 .8% ,回收率前者为 92 .8% ,后者为 91.2 %。线性关系前者为 10~ 160mg/L ,后者为 2 0~ 2 0 0mg/L。 结论 :用气相色谱法测定大鼠血清中的EPA与DHA快速、准确且灵敏度高。

【Abstract】 Objective:To find out,with gas chromatography,the changes in the contents of EPA and DHA in rat serum due to the changes in the composition of food for rats.Methods:10%diethyleneglycol succinate(DEGS) column was chosen.Samples were extracted with ethyl acetate and were methylated with BF3 ether/methanol.Results:The methylation rate of both EPA and DHA was 97.1%~100.8%,and the mean recovery rate of EPA was 92.8%,and of DHA,91.2%,respectively.The linear relationship of EPA was 10~160mg/L serum,and of DHA,20~200mg/L serum.Conclusion:Gas chromatography is a rapid,simple,sensitive and very precise method for the determination of EPA and DHA levels in rat serum.
