bFGF、NGF对高温影响神经细胞存活的保护作用
THE PROTECTION OF bFGF AND NGF ON NEUROEPITHELIAL CELLS FROM HYPERTHERMIA
【摘要】 目的 :研究高温对原代培养神经细胞存活的影响以及bFGF和NGF对高温处理后神经细胞存活的保护作用。方法 :应用神经细胞原代培养、MTT、NSE ELISA测定技术。结果 :①高温处理后神经细胞活性明显降低 ;②外源性bFGF、NGF均不同程度地增加了神经细胞活性 ,其抗体均不同程度地使神经细胞活性降低 ,但二者联合应用神经细胞活性增加不明显 ;③NSE ELISA检测结果与MTT检测结果一致 ,但特异性和敏感性更高。结论 :外源性bFGF和NGF对高温处理后神经细胞存活具有保护作用。
【Abstract】 Objective:To study the effects of hyperthermia on the survival of neuroepithelial cells in primary culture and protections of exogenous bFGF and NGF on them treated with hyperthermia.Methods: Primary culture technique of neurons,MTT and NSE ELISA assays were used.Results: ①Hyperthermia decreased the activities of mitochondrionase of neuroepithelial cells.②Both bFGF and NGF increased the activities of neuroepithelial cells to some extent compared with control group,whereas anti bFGF and anti NGF reduced their effects.Although combination of bFGF and NGF made the activities increase,there was no significant difference compared with control group.③NSE ELISA analysis showed similar result to that of MTT assay,but its specifity and sensitivity were higher.Conclusion:Exogenous bFGF and NGF may protect neuroepithelial cells from hyperthermia influence.
